Product Specific Information
The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat FOXP3 (FORKHEAD BOX P3, SCURFIN, JM2).
FOXP3 is a 49–55 kDa transcriptional regulator identified as the defective gene in scurfy mice. High FOXP3 expression is characteristic of CD4+CD25+ regulatory T cells (Treg).
- Epitope mapping: amino acids 75–125 of mouse FOXP3
- Recommended buffer: Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00)
- Application tested: Intracellular staining and flow cytometric analysis of mouse splenocytes
- Suggested protocol: BestProtocols® – Protocol B: One-step staining for intracellular (nuclear) proteins
Usage note: ≤0.5 µg/test in 100 µL final volume (10⁵–10⁸ cells/test). Optimize concentration for best performance.

FOXP3 (Forkhead box protein 3)
FOXP3 is a transcriptional regulator crucial for immune homeostasis, expressed in CD25+CD4+ regulatory T cells. It is 381 amino acids long and absent in CD8+ T cells.
Defects in FOXP3 cause scurfy phenotype in mice and IPEX syndrome in humans (immune dysfunction, polyendocrinopathy, enteropathy, X-linked).
FOXP3 expression is regulated by p53 in response to DNA damage in human breast and colon cancer cells.
