Product Specific Information

The Sa2-8 monoclonal antibody reacts with mouse CD14, a 53–55 kDa GPI-linked glycoprotein. CD14 acts as a receptor for LPS–LBP complexes and associates with Toll-Like Receptor 4 (TLR4), participating in cellular responses to bacterial LPS. In mice, CD14 is expressed on macrophages and can also be found in serum in a soluble form. The Sa2-8 clone exhibits weak antagonistic activity in NF-κB activation or TNF-α production upon LPS stimulation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of thioglycolate-elicited peritoneal exudate cells. Recommended ≤1 µg/test in 100 µL final volume. Cell count should be empirically determined (10⁵–10⁸ cells/test). Optimize antibody titration for best assay performance.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a multifunctional receptor for lipopolysaccharide (LPS) in complex with LPS-binding protein (LBP). Soluble CD14 (sCD14) exists in serum and contributes to LPS recognition and neutralization. CD14 also binds apoptotic cells and influences allergic, inflammatory, and infectious processes. Alternative splicing produces multiple transcript variants encoding the same protein. CD14 dysfunction is associated with mycobacterium chelonae infection and croup.
