Product Specific Information
- For Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris Buffer with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
TNF alpha (Tumor Necrosis Factor alpha) is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is primarily secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (dimers, trimers, or pentamers) and shows a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout mouse studies indicate a neuroprotective function, and TNF alpha induces tumor necrosis when injected into tumor-bearing mice. It also plays a role in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cell cytolysis, particularly when synergized with interferon-gamma.
TNF alpha has been implicated in autoimmune diseases, insulin resistance, and cancer.
