Target Information
TNF-alpha (Tumor Necrosis Factor alpha) is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is primarily secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-alpha regulates immune responses, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers of two, three, or five noncovalently linked units and shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice have suggested its neuroprotective role and tumor necrosis-inducing activity. TNF-alpha is also involved in immune responses to bacterial, viral, parasitic, and fungal infections, and contributes to tumor cell cytolysis, often synergistically with interferon-gamma. Dysregulation of TNF-alpha is associated with autoimmune diseases, insulin resistance, and cancer.
