Product Specific Information

The SK1 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed as CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on thymocytes, mature T cells, and NK cells, and binds MHC class I molecules, mediating T-cell development and activation through p56lck kinase.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated on normal human peripheral blood cells.
Use at 5 µL (0.06 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein critical for T-cell signal transduction, differentiation, and activation. Expressed mainly on T cells as CD8α/β heterodimers, it acts as a co-receptor with TCR for MHC class I molecules. CD8α also forms homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Upon MHC-I binding, it recruits Lck kinase, leading to cytokine production and cytotoxic T lymphocyte activation, essential for pathogen and tumor cell clearance. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
