Product Specific Information

The SK1 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8α/β) or homodimer (CD8α/α). CD8a is found on most thymocytes and subsets of mature T cells and NK cells. It binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use: 5 µL (0.06 µg)/test in 100 µL final volume with 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL of buffer per 100 µL of cell sample containing 10³–10⁸ cells.
  • NovaFluor conjugates utilize Phiton™ technology for engineered fluorescent signatures minimizing spillover and spread.

Target Information

CD8 is a type I transmembrane glycoprotein of the immunoglobulin family, essential in T cell signal transduction and activation. It is expressed mainly on T cells as a CD8α/β heterodimer and functions as a co-receptor for MHC-I molecules. CD8 also appears as CD8α homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes triggers Lck-mediated signaling leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are critical for pathogen and tumor clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
