Target Information
Adenosine receptors (ARs) are members of the 7-transmembrane domain G-protein-coupled receptor superfamily. Structural, biochemical, and pharmacological analyses have identified four distinct AR subtypes (A1, A2a, A2b, A3). Activation of ARs mediates receptor subtype-specific physiological processes such as cardiac rate regulation, smooth muscle tone, platelet aggregation, inflammation, cell growth and death, and neurotransmission.
The A1 adenosine receptor (A1AR) is a glycoprotein that activates Gi and Go proteins. In intact cells, A1AR activation causes pertussis toxin-sensitive inhibition of adenylyl cyclase and may stimulate phospholipase C, mobilizing intracellular calcium. Activation of potassium channels by A1AR influences cardiovascular function. A1AR is highly expressed in brain regions (cerebellum, hippocampus, thalamus, cortex) and spinal cord, modulating neurotransmitter release. In white adipocytes, A1AR inhibits lipolysis and stimulates glucose uptake. It is also expressed in kidney and testis.
