Eukaryotic cells contain three types of prenyltransferases that attach either a farnesyl group (15 carbons) or a geranylgeranyl group (20 carbons) via thioether linkage to C-terminal cysteine residues in various proteins. These posttranslational modifications help localize proteins to membranes lacking transmembrane domains.

CAAX farnesyltransferase (FTase) transfers a farnesyl group from farnesyl pyrophosphate to cysteine residues at the fourth position from the C-terminus of proteins ending in the CAAX box (C = cysteine, A = usually aliphatic amino acid, X = typically methionine or serine). This enzyme can farnesylate peptides as short as four residues that match the CAAX consensus sequence. The gene for the beta subunit of CAAX farnesyltransferase (FNTB) is located at 14q23–q24, identified through Southern blot hybridization, PCR analyses of human/Chinese hamster somatic cell hybrids, and fluorescence in situ hybridization.
