Product Specific Information

Description:
The HA58 monoclonal antibody reacts with human CD54 (ICAM-1), a 90–110 kDa transmembrane glycoprotein expressed by monocytes, lymphocytes, and endothelial cells. CD54 expression increases upon lymphocyte activation and plays a key role in the inflammatory response through interaction with CD11a.

Applications Reported:
Reported for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometry using normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 is excited with a 405 nm violet laser and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent. Ensure your instrument can detect this fluorochrome.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein with five immunoglobulin superfamily repeats, a transmembrane region, and a cytoplasmic domain. It has seven potential N-linked glycosylation sites and shares sequence homology with ICAM-2 and ICAM-3.
ICAM-1 binds to integrins CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for Rhinovirus. It is expressed on activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, and immune cells such as B and T lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils, with expression increasing during inflammation. Soluble ICAM-1 is detectable in plasma and elevated in inflammatory conditions.
