Target Information

Estrogen Receptor beta (ERβ, NR3A2) is a nuclear hormone receptor protein of approximately 55 kDa, with multiple isoforms ranging from 36–59 kDa. Human ERβ was initially cloned and characterized from testis. Estrogen receptors are ligand-activated transcription factors that, upon binding estrogen, undergo conformational changes enabling dimerization and binding to estrogen response elements (EREs) in DNA. This binding regulates gene transcription through recruitment of coactivator or corepressor proteins.

Two forms of estrogen receptors exist—ERα and ERβ—encoded by separate genes (ESR1 and ESR2). ERβ binds estrogens with similar affinity to ERα and can inhibit ERα activity in reproductive tissues. ERβ is widely expressed in many tissues and functions as a potential tumor suppressor, playing roles in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease.
