Background

This gene belongs to the Tre-2/BUB2/CDC16 domain family. The encoded protein contains two phosphotyrosine-binding domains (PTB1, PTB2), a calmodulin-binding domain (CBD), a Rab-GTPase domain, and multiple AKT phosphomotifs. It regulates insulin-dependent trafficking of the glucose transporter 4 (GLUT4), facilitating glucose uptake into skeletal muscle and adipose tissues. Reduced expression increases GLUT4 at the plasma membrane, indicating its role in intracellular retention under basal conditions. Upon insulin stimulation, AS160 becomes phosphorylated, dissociates from GLUT4 vesicles, increasing GLUT4 surface expression and enhancing glucose transport.
