Cell membrane; single-pass type I membrane protein. Found in perikaryon, growth cone, clathrin-coated pits, early endosome, and cytoplasmic vesicles.
Most APP is localized in intracellular vesicles with only a minor proportion at the cell surface.
During maturation, immature APP (N-glycosylated in the endoplasmic reticulum) moves to the Golgi complex for complete maturation (O-glycosylation and sulfation). After α-secretase cleavage, soluble APP is released extracellularly, and the C-terminal fragment is internalized to endosomes and lysosomes. Some APP accumulates in secretory transport vesicles. 배경 (Background)
This gene encodes a cell surface receptor and transmembrane precursor protein that is cleaved by secretases to form several peptides.
Some peptides are secreted and interact with the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the amyloid plaques characteristic of Alzheimer’s disease.
Certain peptides exhibit antimicrobial activity against bacteria and fungi.
Mutations in this gene are associated with autosomal dominant Alzheimer’s disease and cerebral amyloid angiopathy.
Multiple transcript variants encoding different isoforms have been identified (RefSeq, Aug 2014).
