Background
Mitogen-activated protein kinase (MAPK) signaling cascades include MAPK or extracellular signal-regulated kinase (ERK), MAPK kinase (MKK or MEK), and MAPK kinase kinase (MAPKKK or MEKK).
MAPKKK5 phosphorylates and activates its downstream kinase MKK4, which subsequently activates c-Jun N-terminal kinase (JNK)/stress-activated protein kinase (SAPK).
MAPKKK5 is abundantly expressed in human heart and pancreas, and does not activate MAPK/ERK.
This conserved signaling pathway plays a key role in stress and apoptosis regulation across species from yeast to mammals.
