Target Information
Members of the frizzled gene family encode 7-transmembrane domain proteins that serve as receptors for Wnt signaling proteins. The Frizzled 5 protein is believed to be the receptor for the Wnt5A ligand. It has been reported to be expressed in fetal kidney, fetal and adult liver, fetal lung, and adult pancreas. ESTs have been isolated from bone, liver/spleen, placenta, and prostate libraries. Frizzled 5 was cloned from a retina cDNA library.
Frizzled 5 functions as a receptor for Wnt proteins and is part of the Wnt-Fzd-LRP5-LRP6 complex that triggers β-catenin signaling through aggregation of receptor-ligand complexes. This canonical pathway activates disheveled proteins, inhibits GSK-3 kinase, promotes nuclear accumulation of β-catenin, and activates Wnt target genes. Some family members also signal through PKC and calcium fluxes, possibly integrating with the canonical pathway. Both pathways involve G-protein interactions and may contribute to polarity information transmission during tissue morphogenesis and differentiation. Frizzled 5 also acts as a coreceptor with RYK for Wnt proteins such as WNT1.
