Target Information
COX2 converts arachidonate to prostaglandin H2 (PGH2), a committed step in prostanoid synthesis, including production of inflammatory prostaglandins. The conversion involves a cyclooxygenase (COX) reaction converting arachidonate to prostaglandin G2 (PGG2), followed by a peroxidase reaction reducing PGG2 to PGH2.
COX2 is constitutively expressed in certain tissues (endothelium, kidney, brain) and up-regulated under pathological conditions such as cancer and inflammation. Up-regulation is linked to increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 promotes prostaglandin E2 (PGE2) production, influencing motility, proliferation, and apoptosis resistance. It is naturally inhibited by calcitriol (active Vitamin D). Glucocorticoids repress PTGS2 gene activity in vivo, and COX2 is a known target of NSAIDs like aspirin, which reduce pain and inflammation.
