Target Information
Mammalian serine hydroxymethyltransferase (SHMT)는 사량체(pyridoxal phosphate, PLP)-의존 효소로, serine과 tetrahydrofolate의 가역적 상호 전환을 촉매하여 glycine과 methylenetetrahydrofolate를 생성한다.
이 효소는 세포질(cSHMT, SHMT1)과 미토콘드리아(mSHMT, SHMT2)에 존재하며, cSHMT는 주로 thymidylate 생합성을 위한 one-carbon unit을 공급하고, serine 합성을 통해 methylenetetrahydrofolate pool을 고갈시켜 S-adenosylmethionine(SAM) 합성을 억제한다.
