The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with BCR to mediate signaling, tyrosine phosphorylation, calcium flux, and B cell proliferation.

Each product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.4 µg)/test.

NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates. Add 5 µL per sample or per 100 µL of cell suspension containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures engineered for optimized spectral performance and reduced spillover.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on all peripheral B cells. It is also found on certain myeloid leukemia cells. CD19 acts as a signal-amplifying coreceptor for the B cell receptor (BCR) and regulates B cell proliferation. Mutations lead to hypogammaglobulinemia, while overexpression causes B cell hyperactivity.
