Product Specific Information

The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed on B cells throughout development except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, Leu13, and MHC class II, associating with BCR to mediate signaling, tyrosine phosphorylation, calcium flux, and B cell proliferation. The SJ25C1 and HIB19 antibodies recognize overlapping epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Use at 5 µL (0.4 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Compatibility Note:
NovaFluor dyes are incompatible with DNA-intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining. Add 5 µL per sample or per 100 µL cell suspension for single-color controls. Use in antibody cocktails before combining with cells.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. Expressed on all peripheral B cells and some myeloid leukemia cells. CD19 acts as a signal-amplifying coreceptor for BCR and regulates B cell proliferation and differentiation. Mutations cause hypogammaglobulinemia; overexpression leads to B cell hyperactivity. It is a key marker for B cell lineage and leukemia phenotype studies.
