Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of blood plasma proteins.
It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants (in vitro).
Does not require ATP and maintains partially unfolded proteins for refolding by other chaperones (e.g., HSPA8/HSC70).
Binding to cell surface receptors triggers internalization and degradation via lysosomal or proteasomal pathways.
Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis.
Intracellular isoforms interact with ubiquitin and SCF (SKP1-CUL1-F-box protein) E3 ligase complexes to promote ubiquitination and proteasomal degradation of target proteins (e.g., COMMD1, IKBKB).
Modulates NF-kappa-B transcriptional activity.
Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis.
Plays a role in regulation of cell proliferation.
