Target Information
TNK2 (ACK), also known as activated Cdc42-associated kinase, is a non-receptor tyrosine kinase that functions downstream of Cdc42. It plays a role in tumorigenesis and metastasis, particularly in prostate cancer. TNK2 mediates CDC42-dependent cell migration via phosphorylation of BCAR1 and may contribute to adult synaptic function, plasticity, and brain development. It activates AKT1 by phosphorylating Tyr-176, and phosphorylates AR at Tyr-267 and Tyr-363, promoting recruitment to androgen-responsive enhancers. Additionally, it phosphorylates WWOX (Tyr-287) and MCF2, enhancing GEF activity toward Rho family proteins. TNK2 regulates AXL receptor levels, influences tumor growth, and modulates pro-survival and tumor suppressor pathways.
