Target Information
Phospholipase C gamma (PLCγ) family in mammals includes PLCγ1 and PLCγ2, which share core structural domains and unique regions containing an additional PH domain, two SH2 domains, and one SH3 domain. Upon extracellular stimulation by hormones or growth factors, receptor tyrosine kinases (RTKs) phosphorylate and activate PLCγ. The activated enzyme catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2), generating second messengers inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG).
