Pepsinogens are inactive precursors of pepsin, the major acid protease in the stomach. Pepsin arises from pepsinogen by removal of a 41-amino-acid segment from the N-terminus. It is synthesized in the stomach lining, and hydrochloric acid is required for activation and maintenance of optimal acidity (pH 1–3). Pepsin cleaves peptide bonds involving aromatic amino acids, showing broad specificity. PGA5 belongs to the A1 subfamily of the pepsin family and is the predominant endopeptidase in vertebrate gastric juice. It is inhibited by ovUS-1, a uterine serpin.
