Target Information
DR5 (Apo2, TRAIL-R2, TRICK2, KILLER)는 TRAIL에 의해 유도되는 apoptosis를 매개하는 death domain을 포함한 수용체입니다. DR5는 TNF-receptor superfamily의 구성원으로, intracellular death domain을 포함합니다.
DR5는 TNFSF10/TRAIL/APO-2L에 의해 활성화되어 apoptosis 신호를 전달하며, FADD-deficient mice 연구에서 FADD가 DR5 매개 apoptosis에 필요함이 확인되었습니다.
두 가지 전사 변이체(isoforms)와 하나의 non-coding transcript가 존재합니다. DR5 과발현 시 apoptosis 유도 및 NF-kappa-B 활성화가 일어나며, 림프구와 비장에서 높은 수준으로 발현됩니다.
DR5 기능 이상은 편평세포암(squamous cell carcinoma) 및 diffuse infiltrative lymphocytosis syndrome과 관련이 있습니다.
