Target Information
TRF1 is a negative regulator of telomere length maintenance. Recently, two proteins—tankyrase and Tin2—which bind to mammalian TRF1, have been identified.
Tankyrase has homology to ankyrins and the catalytic domain of poly(ADP-ribose) polymerase (PARP). It localizes to telomeres by binding to TRF1 through its ankyrin repeats and exhibits PARP activity.
ADP-ribosylation of TRF1 reduces its ability to bind telomeric DNA, suggesting that telomere function is regulated by poly(ADP)-ribosylation.
TIN2 was identified as a TRF1-interacting partner via yeast two-hybrid screening. It localizes to telomeres and is essential for proper telomere length regulation. Like TRF1, TIN2 is constitutively expressed, acting to counterbalance telomere elongation by telomerase, though endogenous levels may be low and difficult to visualize by immunostaining.
