Target Information
Glutamate is the main excitatory neurotransmitter in the brain. Initially known to act only on ligand-gated receptor channels (NMDA, AMPA, kainate receptors) involved in fast excitatory synaptic transmission, glutamate is now recognized to regulate enzymes producing second messengers via specific receptors coupled to G-proteins—called metabotropic glutamate receptors (mGluRs).
Group I receptors (mGluR1a, mGluR5) stimulate phospholipase C, increasing phosphoinositide turnover and calcium release from internal stores.
Group II and III receptors (including mGluR2 and mGluR3) inhibit adenylyl cyclase activity.
