Product Specific Information

The HIP1 monoclonal antibody reacts with the human CD42b molecule, a 135 kDa transmembrane protein also known as platelet gpIb.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested for flow cytometric analysis of human platelets.
Recommended use: 5 µL (0.5 µg) per test in a total volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD42b is a platelet activation marker involved in coagulation and platelet aggregation. It interacts with extracellular matrix and adhesion molecules and forms part of the GPIb-IX-V receptor complex critical for platelet-rich thrombus formation. CD42b binds von Willebrand factor (VWF), thrombin, coagulation factors XI and XII, high molecular weight kininogen, TSP-1, integrin Mac-1, and P-selectin. It is expressed on platelets and megakaryocytes in bone marrow. Mutations in CD42b are associated with Bernard-Soulier syndrome and platelet-type von Willebrand disease.
