Target Information
SMCR8는 C9orf72-SMCR8 복합체의 구성 요소로, 구아닌 뉴클레오타이드 교환 인자(GEF) 활성을 가지며 자가포식(autophagy)을 조절합니다. 이 복합체 내에서 C9orf72와 SMCR8은 촉매 서브유닛으로 작용하여 GDP 결합형 RAB8A 및 RAB39B를 GTP 결합형 활성 형태로 전환시켜 오토파고좀 성숙을 촉진합니다. 또한 ATG1/ULK1 키나아제 복합체와 상호작용하여 단백질 키나아제 활성을 억제함으로써 자가포식 개시를 음성 조절합니다. SMCR8은 mTORC1 신호전달을 조절하며, 세포질 내 복합체뿐 아니라 핵에서도 크로마틴과 결합하여 ULK1 및 WIPI2 유전자 발현을 억제합니다.
