Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven transmembrane domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding and fusion. CXCR4 serves as the receptor for CXCL12/SDF-1, increasing intracellular calcium and activating MAPK1/MAPK3 signaling. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels. CXCR4 plays essential roles in hematopoiesis, cardiac and vascular development, and neuronal survival. It binds bacterial LPS and mediates inflammatory responses, including TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of target cells.
