Target Information

Neuron specific enolase (NSE, ENO1, ENO2, ENO3) is an enzyme that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse reaction in gluconeogenesis. NSE is highly stable in biological fluids and can diffuse into the extracellular medium and cerebrospinal fluid (CSF) when neuronal membranes are damaged.

NSE is one of three mammalian enolases, also known as ENO1, ENO2, and ENO3 (or enolase α, β, and γ). The α-subunit is expressed in most tissues, the β-subunit in muscle, and the γ-subunit mainly in neurons and neuroendocrine cells. Co-expression of NSE and chromogranin A is common in neuroendocrine neoplasms.

Because neurons require high energy, they are rich in glycolytic enzymes such as GAPDH and NSE. Antibodies to NSE are useful for identifying neuronal cell bodies, developing neuronal lineages, and neuroendocrine cells. Release of NSE from damaged neurons into CSF and blood serves as a biomarker for neuronal injury.

Clinically, NSE is a sensitive marker for neuronal damage in disorders such as stroke, hypoxic brain damage, status epilepticus, Creutzfeldt-Jakob disease, and herpetic encephalitis. Elevated plasma NSE levels are also observed in neoplasias including pediatric neuroblastoma and small cell lung cancer.
