Target Information
Pantothenate kinase (PANK2) is a key regulatory enzyme in coenzyme A (CoA) biosynthesis, catalyzing the cytosolic phosphorylation of pantothenate (vitamin B5), N-pantothenoylcysteine, and pantetheine. CoA functions as a major acyl carrier, essential for intermediary and fatty acid metabolism.
Mutations in PANK2 cause pantothenate kinase-associated neurodegeneration (PKAN), formerly known as Hallervorden-Spatz syndrome (HSS)*—an autosomal recessive neurodegenerative disorder characterized by iron accumulation in the brain. PANK2 mutations are also linked to *hypoprebetalipoproteinemia, acanthocytosis, retinitis pigmentosa, and pallidal degeneration (HARP).
