Biological Background
The hnRNPs are RNA-binding proteins that form complexes with heterogeneous nuclear RNA (hnRNA) and associate with pre-mRNAs in the nucleus. These complexes are involved in pre-mRNA processing, mRNA metabolism, and transport.
Although all hnRNPs are present in the nucleus, evidence suggests that they shuttle between the nucleus and cytoplasm. hnRNP proteins possess distinct nucleic acid-binding properties.
The hnRNP A1 protein contains two quasi-RRM domains that bind RNA. This abundant core protein, along with other hnRNP proteins, is exported from the nucleus—likely bound to mRNA—and rapidly re-imported. hnRNP A1 is essential for packaging pre-mRNA into hnRNP particles, transporting poly(A)+ mRNA from nucleus to cytoplasm, and may influence splice site selection.
