This expression vector contains the CMV promoter for the expression of genes in mammalian cells. The plasmid also includes the internal ribosome entry site (IRES) from Foot and Mouth Disease Virus (FMDV) that enables two proteins to be produced from the same mRNA. In this vector there is a secreted luciferase that we have developed immediately downstream from the IRES that allows plasmid activity to be measured in the supernatant of mammalian cultures. Using IRES expression systems can often produce low levels of expression particularly in some cell types for this reason we can provide other plasmids with higher levels of reporter gene expression such as pSF-CMV-PGK-iLumena or pSF-CMV-Ub-iLumena.

Promoter Expression Level: Promoter and IRES Expression Level: This plasmid contains the mammalian CMV promoter to drive gene expression. We have tested all of our mammalian promoters in a range of cell types and CMV is consistently the strongest in those we have studied. However there are many reports of the CMV promoter demonstrating silencing by methylation in long-term culture. The IRES that is used to drive iLumena secreted luciferase expression from the CMV promoter is considerably weaker than the CMV promoter and typically yields >20 fold lower protein levels.
