Target Information
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) is an inhibitory coreceptor involved in regulation of T cell and B cell signaling through dual immunoreceptor tyrosine-based inhibitory motifs (ITIMs). These motifs, upon phosphorylation by associated kinases, provide docking sites for protein-tyrosine phosphatases.
CD31 is ubiquitously expressed within the vascular compartment, primarily at junctions between adjacent endothelial cells. Its N-terminal Ig-like domain mediates homophilic binding, playing a crucial role in cell-cell interactions.
This multifunctional molecule modulates:
- Integrin-mediated cell adhesion
- Transendothelial migration
- Angiogenesis and apoptosis
- Negative regulation of immunoreceptor signaling
- Autoimmunity and macrophage phagocytosis
- IgE-mediated anaphylaxis and thrombosis
