Target Information

ZO-3 is a member of the membrane-associated guanylate kinase (MAGUK) family and is considered a tight junction-associated protein. Tight junctions create intercellular boundaries between epithelial and endothelial cells.
ZO-3 is believed to have both structural and signaling roles and is defined by three protein-protein interaction modules: PDZ, SH3, and GuK domains.
ZO-1 forms complexes with ZO-2 or ZO-3, and these proteins can associate with claudin, occludin, and F-actin at tight junction strands, linking the actin cytoskeleton with the junction.
ZO-1 expression is significantly reduced in many breast cancer lines. ZO-2 and ZO-3 are ubiquitously expressed in epithelial tight junctions; unlike ZO-1, ZO-2 is not expressed in non-epithelial tissue.
