Target Information
Folylpoly-gamma-glutamate synthetase (FPGS) catalyzes the addition of several equivalents of glutamic acid to the gamma-carboxyl group in the side chain of folate cofactors and analogs.
FPGS has three major functions in mammalian folate homeostasis:
- Polyglutamation prevents efflux of folate cofactors from the cell.
- It increases binding of folate cofactors to enzymes involved in folate interconversion and biosynthesis.
- It enables accumulation of folates in mitochondria required for glycine synthesis.
FPGS is widely expressed in human tumors and is associated with cell proliferation or lack of differentiation. Its function enhances the efficiency of folate antimetabolites by maintaining inhibition of target enzymes over multiple cell generations.
