Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, binding glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures optimized for minimal spillover and spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor composed of:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

It is constitutively expressed on all leukocyte classes, including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
