Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells and binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤ 0.2 µg/test, 100 µL final volume)

Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Conjugates:
Based on Phiton™ technology utilizing novel nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.
Excitation: 509 nm; Emission: 614 nm; Laser: 488 nm (Blue)

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein member of the selectin family of adhesion receptors. It includes:

• Aminoterminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

CD62L is constitutively expressed on leukocytes including lymphocytes (except memory T cells), monocytes, and polymorphonuclear cells.
