Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was originally identified as a surface antigen on Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1.
The ligand for CD30 is CD30L (CD153), and their interaction mediates various cellular responses including proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas, acting as a costimulatory molecule in thymic negative selection. It is expressed on Hodgkin’s and Reed-Sternberg cells, some non-Hodgkin’s lymphomas (including Burkitt’s lymphoma), virus-infected T and B cells, and activated normal T and B cells.

In T cells, CD30 expression is found on subsets producing Th2-type cytokines and on CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL-4 receptor. The soluble form (sCD30) serves as a marker of Th2 immune response.
TRAF2 and TRAF5 interact with CD30 to activate NF-κB signaling. CD30 acts as a positive regulator of apoptosis, limiting autoreactive CD8 effector T cell proliferation and contributing to immune regulation.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
