Target Information
Survivin (IAP4)은 세포자멸 억제 단백질(Inhibitors of Apoptosis Protein, IAP) 패밀리의 가장 작은 구성원입니다. 이 단백질들은 세포 신호전달, 세포분열, 대사 등 다양한 세포 기능에 관여하며, 대부분의 암세포에서 발현되지만 정상 조직에서는 거의 발현되지 않습니다. Survivin은 단일 BIR 도메인을 가지며 RING finger 도메인을 결여하고 있습니다. 태아 발달기와 종양에서 발현이 높고, 성체 조직에서는 낮은 수준을 보입니다.
세포주기 G2/M 단계에서 주기적으로 발현되며, 유사분열 초기에는 미세소관과 결합하여 세포분열을 조절합니다. Survivin-미세소관 상호작용이 방해되면 항-아포토시스 기능이 손실되고 caspase-3 활성이 증가하여 세포사멸이 촉진됩니다. 또한, 뇌 손상 후 성체 랫트의 신경조직(성상세포 및 일부 뉴런)에서 높은 발현이 관찰되며, NeuN 및 PCNA와의 공발현이 보고되었습니다. Survivin은 신경세포의 증식 반응 조절에도 관여할 수 있습니다. 이 유전자는 최소 4개의 전사 변이체를 가지며, 그중 3개의 전장 서열이 확인되었습니다. 암세포에서의 과발현은 세포주기 조절 이상과 관련된 것으로 알려져 있습니다.
