Target Information

Voltage-gated potassium (Kv) channels are essential components of potassium currents in the heart and central nervous system. In cardiac tissue, Kv4 channels participate in the repolarization phase of the action potential, while in the brain, they prevent reverse propagation of action potentials.
KChIPs (Kv channel interacting proteins) are calcium-binding proteins that associate with Kv4 channels to regulate their function. There are three known KChIPs:

  • KChIP1: expressed in the brain
  • KChIP2: expressed in brain, heart, and lung (three isoforms: a, b, c)
  • KChIP3: also known as calsenilin or DREAM, expressed in brain and testis

KChIP3 not only modulates Kv4 currents but also acts as a transcriptional regulator by binding to the regulatory DRE and repressing transcription of early-response genes such as c-fos. It is also associated with presenilins and implicated in beta-amyloid formation.
