Applied Biosystems™ TaqMan™ Copy Number Reference Assay, Human TERT
Human TaqMan™ Copy Number Reference Assays are designed for duplex real-time PCR reactions in combination with human TaqMan™ Copy Number Assays to detect and measure copy number variations (CNVs) and smaller genomic regions.
Two options for endogenous human reference genes are available: RNase P and TERT.
These assays target unique genomic sequences from the reference genome assembly (Build 37/hg19) and are essential for relative quantitation of copy number targets.
TaqMan™ Copy Number Assays quantify the gene of interest and normalize to an endogenous reference gene known to exist in two copies in a diploid genome.
Note: TaqMan™ Copy Number Reference Assays are species specific.
TERT: The Alternative Reference Gene Option
TaqMan™ Copy Number Reference Assays include a VIC™ dye–labeled TAMRA™ probe and sequence-specific forward and reverse primers.
The assays are not primer-limited.
RNase P Reference Assay (Standard)
- Detects Ribonuclease P RNA component H1 (H1RNA) gene (RPPH1) on chromosome 14, cytoband 14q11.2
- Location: chr.14:20811565 (NCBI build 37)
- Amplicon: 87 bp within the single exon RPPH1 gene
TERT Reference Assay (Alternative)
- Targets the telomerase reverse transcriptase (TERT) gene on chromosome 5, cytoband 5p15.33
- Location: chr.5:1253373 (NCBI build 37)
- Amplicon: 88 bp within exon 16 of the TERT gene
- Recommended when RNase P assay performs poorly due to chromosomal aberrations or other sample issues
Simplest Copy Number Analysis Workflow
Combine the test assay (FAM™ dye–labeled), reference assay (VIC™ dye–labeled), sample DNA, and TaqMan™ Master Mix.
Run on an Applied Biosystems™ real-time PCR system using the standard TaqMan™ Copy Number Assay protocol.
Total setup and primary analysis typically take 3–4 hours, including a ~2-hour PCR run.
