Target Information
Ubiquitin-mediated proteolysis involves the transfer of ubiquitin (Ub) to lysine residues on target proteins, promoting their degradation via the 26S proteasome. Ub-fusions are processed by Ub-specific proteases (UBps) or by the Ub-fusion degradation (UFD) pathway. The UFD1 protein, first identified in S. cerevisiae, has a human homolog UFD1L. In vitro, UFD1 attenuates degradation of Ub-fusions with Gly76 substitutions through selective multiubiquitination. Mutations in UFD1 are linked to CATCH22 syndrome, associated with cardiac defects, cleft palate, and hypocalcemia, indicating a role in developmental processes.
