Target Information

NLK is a serine/threonine kinase involved in signal transduction and cellular communication. It acts as a positive effector of the non-canonical Wnt signaling pathway downstream of WNT5A, MAP3K7/TAK1, and HIPK2. Activation leads to phosphorylation of the histone methyltransferase SETDB1, forming a complex that interacts with PPARG and silences transcription of PPARG target genes, thereby inhibiting adipogenesis and promoting osteoblastogenesis in mesenchymal stem cells.

NLK negatively regulates:

  • Canonical Wnt/β-catenin signaling by phosphorylating TCF7L2/TCF4 and LEF1.
  • Notch signaling by phosphorylating NOTCH1.
  • MYB family transcription factors.
