Target Information
C5b-9 membrane attack complexes are assembled from five precursor molecules in serum. Proteolytic cleavage of C5 by C5 convertase generates C5b, which initiates assembly of the C5b-9 complex. The final step involves polymerization of C9, allowing insertion of the complex into the cell membrane. During C5b-8 formation and C9 polymerization, neoantigens unique to C5b-9 appear. Complement regulatory proteins such as CD59 and complement S-protein can prevent membrane insertion. The SC5b-9 complex, unable to attach to cells, is cytolytically inactive. C5b-9 plays a role in chronic proteinuric renal disease progression by mediating continuous tubulointerstitial damage, which can be mitigated when C5b-9 formation is inhibited.
