Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein belonging to the immunoglobulin receptor family. It plays a crucial role in T-cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer (CD8α and CD8β) that acts as a co-receptor with the T-cell receptor (TCR) for MHC class I molecules. In contrast, CD4 serves as a co-receptor for MHC class II molecules.

CD8α is essential for MHC-I binding and is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers. Engagement of MHC-I/peptide complexes on antigen-presenting cells triggers recruitment of Lck kinase, leading to lymphokine production, cell motility, and cytotoxic T lymphocyte (CTL) activation. Activated CTLs are key players in eliminating pathogens and tumor cells. Differentiation of naïve CD8+ T cells into CTLs is enhanced by cytokines such as IL-2, IL-12, and TGF-β1.
