Product Specific Information

The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains ITIM motifs and plays a key role in peripheral tolerance and autoimmune disease. It is expressed mainly on activated T and B lymphocytes. PD-1 ligands include PD-L1 (B7-H1) and PD-L2 (B7-DC), which show overlapping functions and expression on normal and activated antigen-presenting cells.

Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product # 11-9969).

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.5 µg) per 100 µL test volume. Cell number: 10⁵–10⁸ cells/test.

Brilliant Violet™ 786 (BV786): Tandem dye emitting at 786 nm, for use with 405 nm violet laser-equipped cytometers. Ensure instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54). Store fixed samples up to 3 days at 4°C in dark.
Not compatible with methanol-based fixation.

Target Information

PD-1 (Programmed Death-1, PDCD-1) is an inhibitory receptor expressed on activated T cells. It binds ligands PD-L1 and PD-L2 on antigen-presenting cells, transmitting inhibitory signals that limit T-cell activation in the absence of danger signals. PD-1 deficiency leads to hyperactivated immune responses and autoimmune disease. Although predicted at 55 kDa, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
