Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human and canine Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is present during the active phases of the cell cycle (G1, S, G2, and mitosis) but absent in resting cells (G0). It localizes to the nucleus during interphase and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins such as CBX3.

Ki-67 expression serves as a proliferation marker in tumor cell studies, indicating the fraction of proliferating cells within a tumor population.

Applications Reported

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested

  • Pre-diluted and tested using intracellular staining of stimulated human peripheral blood cells with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
  • Recommended use: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information

  • Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is designed for cytometers with a 355 nm UV laser.
  • Use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409-75) when combining multiple polymer dye-conjugated antibodies.
  • Light sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation: Compatible with IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark.
  • Methanol compatibility: Suitable for short-term methanol-based fixation (≤1 hour).

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin regions and shows cell cycle–dependent localization. Ki-67 is widely used as a cell proliferation marker, as it is absent in G0 cells but increases through mitosis.
In tumor diagnostics, the Ki-67 index correlates strongly with tumor grade and serves as a reliable indicator of proliferative activity. It is also used as a neuronal marker for cell cycling and proliferation.
