Target Information

ADRBK1 (GRK2) is a serine/threonine kinase that phosphorylates G protein-coupled receptors (GPCRs). GRK2 (83 kDa) is ubiquitously expressed in mammals and plays a key role in regulating GPCR signaling. Upon ligand binding, GPCRs are phosphorylated by GRKs, leading to internalization mediated by beta-arrestin-1 and activation of Ras-Raf-ERK1/2 pathways. GRK2 activity is regulated by phosphorylation (PKC, Src, ERK1/2) and protein interactions. ERK phosphorylates GRK2 on serine 670, providing negative feedback regulation.
