Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (FcγRIII). CD16 exists as two isoforms, FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, NK cells
• FcγRIIIB: Detected on neutrophils (GPI-anchored form)

Binding of IgG activates signal transduction pathways leading to ADCC, phagocytosis, cytokine release, and antigen presentation.

Based on cross-blocking studies, 3G8 recognizes the same epitope as CB16, distinct from B73.1.
Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells (5 µL/test).
A test is defined as staining 10⁵–10⁸ cells in 100 µL volume.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance, ADCC, and viral infection enhancement.

• Gene: FCGR3A (similar to FCGR3B on chromosome 1)
• Expression:
  • FCGR3A on NK cells (transmembrane form)
  • FCGR3B on neutrophils (PI-anchored form)
• Mutations linked to Immunodeficiency 20, Herpes Zoster, and SLE susceptibility.
• Alternative splicing yields multiple isoforms.
