Target Information

Prostaglandins are produced by the metabolism of arachidonic acid. Prostaglandin E2 (PGE2) is one of the five physiologically significant prostanoids. Its wide range of physiologic and pharmacologic effects are mediated through binding to Prostaglandin E2 receptors (EP1, EP2, EP3, and EP4). These effects span immune, endocrine, cardiovascular, renal, and reproductive systems, as well as smooth muscle regulation.

PGE2 is abundant in the brain, where it plays a role in neural functions and inflammation mediation. EP2 receptors are G-protein coupled receptors (358 amino acids) that stimulate adenylyl cyclase via Gs coupling and do not undergo PGE2-induced internalization. EP2 receptors are mainly localized in lung and placental tissues and smooth muscle, contributing to contraction and relaxation mechanisms.
